Spectrophotometric measurement of hexokinase and phosphohexokinase activity.

چکیده
The methods available for the quantitative assay of these enzymes are based on phosphate transfer determinations and pH change of the reaction mixture. They require large quantities of adenosine triphosphate and, for estimation in crude tissue extracts, the addition of inhibitors such as iodoacetate and sodium fluoride (7). The purpose of this paper is to report a simple and rapid spectrophotometric method by which the rates of phosphorylation by the enzymes hexokinase and phosphohexokinase can be measured. Recently, spectrophotometric tests for the other principal enzymes of the glycolytic system of carbohydrate breakdown have been developed, mainly by Warburg and his collaborators. These methods should prove useful tools in the study of disturbances of carbohydrate metabolism, such as those described for tumor tissues. It should be emphasized, however, that several of them are not suitable for crude tissue extracts and considerable modification may prove necessary to adapt the methods for such purposes.

متن کاملRelation of Iron Salts to Inhibition of Glycolysis by Theiler Fa Virus of Mouse Encephalomyelitis
1. Brain homogenates of mice infected with the Theiler FA strain of mouse encephalomyelitis virus show marked inhibition of glucose phosphorylation. 2. A similar effect can be obtained by incubating normal brain homogenates with small amounts of ferrous sulfate. 3. Partially purified preparations of Theiler FA virus contain iron in amounts corresponding to their inhibitory effect on brain glyco...

Background & Aim: Hexokinase and pyruvate kinase are two regulatory enzymes of glycolytic pathway in erythrocytes. Increasing evidence suggests that cigarette smoking which produces free radicals and oxidative stress can cause damage to body macromolecules such as proteins and enzymes. The aim of the present study was to investigate the susceptibility of key enzymes of erythrocytes glycolyt...
متن کاملMicrocalorimetric determination of glucose in reference samples of serum.
Ireport a detailed series of microcalorimetric measurement of glucose concentrations in five reference samples of serum. The method utilized (a) calibration in the actual medium of analysis and (b) a correction for interferences owing to the nonspecificity of the enzyme hexokinase. The microcalorimetric results are compared with the tentative results of analyses obtained by the spectrophotometr...
متن کاملThe role of complex metal ions in the yeast hexokinase reaction.
It is known that many of the enzyme systems which ut,ilize ATPl are activated by inorganic cations. In most instances a divalent cation is required (2) and in some cases monovalent cations are also required (3-5). These effects have usually been assumed to be due to activation of the enzyme by the cation. The demonstration that many of these cations react with ATP to form complex ions (6-10) ma...
